TLR4 dependent generation of a novel bone marrow-derived inflammatory monocyte population required for elicitation of skin contact hypersensitivity responses

چکیده

Abstract Contact hypersensitivity (CHS) is a CD8 T cell mediated response to hapten skin sensitization and challenge. Hapten-primed infiltration into the challenge site during elicitation of CHS requires prior activation novel population Gr-1 +CXCR2 +cells expressing FasL perforin. The goal these studies was further characterize this myeloid-derived cells determine mechanisms generating in bone marrow CHS. myeloid were purified from sensitized mice 2 hrs after by depletion CD3 +, B220 NK1 F4/80 class II MHC +and Ly6G +cells. Flow cytometry analyses indicate >90% remaining CD11c Ly6C CCR2 This at low numbers naïve but increased peak hrs. WT Rag −/−mice T-cell depleted mice. Generation absent challenged TLR4-deficient inhibited treatment with anti-TNFa mAb time Whereas generation neutrophils anti-G-CSF antibody, following These results that marrow-derived perforin inflammation TNFa TLR4 signaling occurs independently activation.

Recovery of Contact Hypersensitivity Responses Following Murine Bone Marrow Transplantation

Bone marrow transplantation (BMT) is often followed by significant morbidity and mortality due to protracted immunodeficiency. We have hypothesized that the bone marrow-ablative regimen may delay the recovery of normal immune function following transplantation by impairing the interaction of host endothelial cells with circulating graftderived lymphocytes. This report compares the relative effe...

Neutrophils are required for both the sensitization and elicitation phase of contact hypersensitivity

Allergic contact dermatitis and its animal model, contact hypersensitivity (CHS), are T cell-mediated inflammatory skin diseases induced by contact allergens. Though numerous cellular and molecular players are known, the mechanism of chemical-induced sensitization remains poorly understood. Here, we identify neutrophils as crucial players in the sensitization phase of CHS. Genetic deficiency of...

A Method for Generation of Bone Marrow-Derived Macrophages from Cryopreserved Mouse Bone Marrow Cells

The broad use of transgenic and gene-targeted mice has established bone marrow-derived macrophages (BMDM) as important mammalian host cells for investigation of the macrophages biology. Over the last decade, extensive research has been done to determine how to freeze and store viable hematopoietic human cells; however, there is no information regarding generation of BMDM from frozen murine bone...

Generation of Mature Monocyte-Derived Dendritic Cells in the Presence of Heparin and Monocyte Conditioned Medium: Phenotypic and Functional Comparison

Background: Dendritic cells (DC) induce tumor or pathogen-specific T cell responses in humans. Several laboratories have developed culture systems, including maturation factors for human DC from peripheral blood monocytes. We comprehensively compared standard maturation stimulus, an autologous monocyte-conditioned medium (MCM), with heparin for their ability to promote uniformly mature DC that ...
